Is it possible to lock the screen orientation of a touchkit application?

AFAIK there is no way to do that in any web application yet. There is
an API draft
, but I don’t know a browser that would actually support this.
You could try to simulate that with some sort of transformations. Still screen would rotate, but you could “rotate it back” with css3 rotation. But if you are looking for a perfect - stop looking.
